Take Highway 427 South to QEW Hamilton to QEW Niagara. Exit onto Highway 420, go through the first set of lights (Stanley Ave) and continue on Falls Ave. Take a right turn onto Macdonald Ave, a left turn onto Centre St, and a right turn onto Ellen Ave, the Hotel is located on the right hand side.
From Peace Bridge:

From Rainbow Bridge:
